The genus-specific primers for Bifidobacterium sp. have been developed according to the available nucleotide sequences from international databases. Primer specificity was estimated with use of DNA of different bacteria representatives. There were obtained no false-positive results with bacteria from the genera different from Bifidobacterium. With every used DNA of bifidobacteria isolates certain positive result has been shown. For both, pure culture of Bifidobacterium spp. and a mixture containing bifidobacteria, bright positive bands on the electrophoresis appeared.
In comparison with reference genus-specific primer pair Bif662 / Bif164 no considerable differences has been shown, that proves xfp-F and xfp-R primers to be reliable for bifidobacteria to be detected on genus leve.

The specific metabolic pathway provides bifidobacteria with unique enzyme which allows detection of those bacteria with biochemical as well as molecular methods. The Xfp enzyme has not been found in any of lactic acid bacteria. The xfp gene is obviously better target for identification of
bifidobacteria at genus level and can be used instead of the 16S rRNA as we used to. 2. The specificity of the primers to bifidobacteria was estimated with use DNA of 9 strains: 3 type strains and 6 confirmed to be bifidobacteria with sequence of 16S rRNA gene. Positive results were
received to all samples. 3. Absence of false-positive results were verified by use of DNA of 24 strains of other lactic acid bacteria and close genera, including Lactobacillus, Lactococcus, Sporolactobacillus, Leuconostoc, Gardnerella. No positive results were obtained.
